Without diving too deep into all of this, I don't think that hub can support triple 4k high refresh rate monitors so you'll be running multiple cables. The hub shouldn't introduce noticeable latency for your keyboard/mouse, but I'm no expert on thunderbolt. I think everything else is derivative of the first point about the hub.
Just do what I did and put the computer in the room next to yours and then make a nice size hole between the two rooms. Run all your cables through the wall. My computer is on one side of the wall and my desk is on the other side so the cables didn’t even have to be that long.
It’s amazing in the summer since all that hot air isn’t blasting into the room
I've done exactly that in my old house, was great and really easy. However, that won't work in my current house.
