Tome Mods with on-kill conditions do not require the Tome weapon to make the killing blow. Instead it has to damage an enemy at least once to mark it with the Tome effect and then be the held weapon when marked enemies are killed by an other source, such as Warframe abilities or companions.
Notice the bolded text. Basically you have to holding the grimoire most of the time to make use of the mod.
This mod will work if you hit an enemy with a grimoire and then kill the enemy with abilities. (Except for exalted weapon) Thus, you have a way to armor strip for many frames, although the long cooldown of the alternative attack and the small area of the regular attack greatly complicate this method of use.
Multishot and punch-through seemed to help a bit. But with fast killing frame abilities it's at least a bit useful in high level areas; pick a corridor enemies swarm from and as the effect starts to stack it makes each following enemy easier to kill until you hit equilibrium with spawn rate.
Thanks for the clarification guys, comments further below had me a bit confused on the triggering conditions (Grimoire kills / assists). So implicitly you're saying it does stack. Cool.
Does this accumulate/stack? Or just a one time de-buff as described?
One time debuff, but stacks with itself if you trigger it multiple times - additively for armor, and using current shields for shields as far as I can tell.
(So 20 kills around an Acolyte completely removes its armor)
It's not a buff/dubuff. Permanent removal.

This mod is stupid strong once you get it rolling in Survival/Defense type missions. I wouldn't take it into exterminates though.
Where Grimoire can actually kill something, you don't really need any armor/shield strip.
Most weapon can kill better than Grimoire,
Maybe below 80. But at level cap, most weapons don't cream enemies with an up to 425m affinity range armor strip while giving you energy. Grimoire is goated. Yall just don't know how to build lol
What's the point of playing at level cap? You get extra rewards? Or is it just a huge waste of time?
....Yeah saying specifically level cap is weird, but the more important aspect is that it's completely possible to kill high-level enemies with the grimoire, especially with the help of warframe abilities(on a weapon that buffs them, who would of thought?) And that's before the reminder that it counts assist kills as long as you have the book equipped.
Saying 425M is similarly weird. I'm not even sure enemies spawn that far in the majority of content. Realistically you'd only need one fosfor for the 200M, and the +25M from Vazarin is definitely not going to make a meaningful difference if you're using them in the first place.
A few things:
1) This debuff is only applied when the Grimoire kills the enemy.
2) This debuff is a traditional debuff; Simply when you kill an enemy within Affinity Range is reduces Armor/Shields by all enemies within 50m of you. If a new enemy spawns in it would not have this debuff. It is not an aura centered around you, it's a debuff that happens when you kill something with the book.
Affinity range is 45m. 60 while using Vazarin with the node unlocked.

And Vazarin adds 25 meters, which would make it 75 maximum.
Fosfors make it effectively global
Attainable on rotation C Munio mirror defense only although supposedly they are offered in cash shop for plat and of course can be traded.
Do we know if this only affects kills with the Grimoire? Language isn't super clear on that.

Not anymore, the patch makes clear now that only get hit by Grimoire once to mark the enemy with this is enough, you can kill marked enemies with any method you wishes.
